dc.description.abstractA Leishmaniose é uma doença parasitária emergente em algumas regiões do Brasil causada por protozoário do gênero Leishmania. No ano de 2002 foi notificado o primeiro caso humano autóctone de Leishmaniose Tegumentar Americana (LTA) no município de Porto Alegre/RS, Bairro Lomba do Pinheiro. No meio urbano, o cão tem papel importante, servindo como fonte de infecção e reservatório do protozoário. Este estudo teve como objetivo avaliar o emprego de ELISA e PCR no diagnóstico da LTA em cães domiciliados na Estrada do Rincão, Bairro Lomba do Pinheiro, Porto Alegre. JESUS (2006) coletou e submeteu 200 amostras de soro de cães oriundos desta área de risco à Imunofluorescência Indireta (IFI) encontrando uma soroprevalência de 3,5% (7/200). Em nosso estudo, empregaram-se o ensaio imunoenzimático para diagnóstico da Leishmaniose Visceral Canina, kit Biomanguinhos/FIOCRUZ, com 61 (30,5%) dos 200 soros caninos sororeagentes e nestes foi realizada a reação em cadeia da polimerase (PCR) na qual apenas a amostra 12C de uma fêmea, sem raça definida, com quatro anos de idade, replicou o DNA de Leishmania spp. Utilizando o Teste Exato de Fisher para a análise estatística quanto à idade, sexo e raça, observou-se que houve associação significativa com a positividade em ELISA e a idade dos cães (p=0,0037), enquanto que quanto ao sexo (p=0,0687) e raça (p=0,1261) não encontramos associação significativa na positividade em ELISA. O Teste de McNemar`s foi aplicado aos resultados obtidos pelas técnicas de ELISA e IFI evidenciando uma diferença muito significativa entre as duas técnicas (p<0,0001). A identificação de cães soropositivos, sem sintomatologia, em área de baixa endemicidade no município de Porto Alegre, com casos humanos autóctones, serve como alerta e ponto de partida para mais investigações sobre a epidemiologia da LTA no RS e também sobre o emprego de outras técnicas de diagnóstico mais sensíveis.pt_BR

In the year of 2002 it was notify the first autochthon human case of American Tegumentary Leishmaniasis (ATL) in the city of Porto Alegre /RS, Bairro Lomba do Pinheiro. In the urban way, the dog has important paper, serving as source of infection and reservoir of the protozoa. This study it had as objective to evaluate the job of ELISA and PCR in the diagnosis of the ATL in dogs domiciliated in the Road of the Rincão, Bairro Lomba do Pinheiro, Porto Alegre. In our study, they had used for diagnosis Canine Visceral Leishmaniasis, kit Biomanguinhos/FIOCRUZ, with 61 (30.5%) positives samples of the 200 canine serum and in these the reaction in chain of polimerase in which only the sample 12C of a female, without defined race, with four years of age, presented the response of the DNA of the Leishmania genus in the PCR. Using the Accurate Test of Fisher for the analysis statistics how much to the age, sex and race, it was observed that it had significant association it enters the positividade in ELISA and the age of the dogs (p=0,0037), whereas how much to the sex (p=0,0687) and race (p=0,1261) we do not find significant association with the positividade in ELISA. The Test of McNemar `s was applied to the results gotten for the techniques of ELISA and IFI having evidenced a significant difference very between the two techniques (p< 0,0001). The identification of soropositives dogs, without symptons, in area of low endemicity in the city of Porto Alegre, with autochthon human case, also serves as alert and starting point for more inquiries on the epidemiology of the ATL in the RS and on the job of others techniques of more sensible diagnosis.en
