dc.description.abstractEm ratos, a amígdala medial póstero-dorsal (AMePD) é uma região sexualmente dimórfica e está envolvida com o controle da reprodução. Os hormônios gonadais masculinos e femininos possuem um papel epigenético importante sobre a morfologia e a função neuro-glial na AMePD. Nesse trabalho dois grandes tópicos inéditos e pertinentes na literatura para o entendimento das bases celulares de funcionamento da AMePD foram estudados: (1) a atividade da NADPHdiaforase (NADPH-d) e o número de neurônios NADPH-d-positivos de ratos adultos, fêmeas ao longo do ciclo estral (diestro, proestro, estro e metaestro) ou após ovariectomia (OVX) seguida por terapia substitutiva consistindo em veículo/óleo (controle), somente estradiol (BE), estradiol e progesterona (BE+P) ou somente progesterona (P) e, (2) a modificação do volume somático e nuclear de neurônios da AMePD de ratas OVX adultas submetidas ao mesmo tratamento substitutivo, com o objetivo de revelar possíveis influências dos hormônios gonadais sobre esses parâmetros morfológicos. A histoquímica para a NADPH-d foi utilizada para estudar a densidade regional e neuronal da atividade da NADPH-d, seguida por uma análise semi-quantitativa utilizando-se a densitometria óptica. Todos os animais foram submetidos ao mesmo protocolo experimental para obtenção dos resultados. Com base nisso, machos apresentaram maior densidade óptica regional e neuronal quando comparados as fêmeas ao longo do ciclo estral, com exceção da fase de diestro (P < 0,01). Nenhuma diferença foi encontrada nesses dois mesmos parâmetros durante o ciclo ovariano (P > 0,05). Quanto ao número de neurônios NADPH-dpositivos, não foram encontradas diferenças estatisticamente significativas entre machos e fêmeas, estas ao longo do ciclo estral (P > 0,05). Como os hormônios gonadais são capazes de gerar respostas supra-fisiológicas distintas daquelas observadas durante o ciclo ovariano normal na AMePD, foram testados também os efeitos da OVX nesses parâmetros. Não houve diferença estatisticamente significativa na densidade óptica regional, na densidade óptica neuronal e no número de neurônios NADPH-d-positivos quando se comparam fêmeas OVX tratadas com óleo ou com as três diferentes terapias hormonais (P = 0,07, P = 0,18 e P = 0,95, respectivamente). Para medir os volumes somático e nuclear dos neurônios da AMePD, uma semana após a OVX e ao final do tratamento hormonal, os encéfalos foram cortados em secções semi-finas (1 m) e corados com azul de toluidina a 1% para a estimação estereológica, utilizando-se o método de Cavalieri associado à técnica de contagem de pontos. Tanto o volume somático quanto o neuronal apresentaram diferença estatisticamente significante quando comparados os dados entre as fêmeas OVX tratadas com óleo, BE, BE+P ou somente P (P < 0,05). Ou seja, ratas OVX tratadas com BE+P apresentaram maior volume somático e nuclear quando comparadas às fêmeas OVX tratadas com óleo (P < 0,01) ou somente com BE (P < 0,01). Além disso, fêmeas OVX tratadas somente com P apresentaram maior volume somático e nuclear quando comparadas às fêmeas OVX tratadas com óleo (P < 0,05). Esses resultados sugerem que a atividade da NADPH-d é diferente entre os sexos, mas em fêmeas não é afetada pelas mudanças nos níveis de hormônios gonadais em condições fisiológicas ou suprafisiológicas. Além disso, os volumes somáticos e neuronais podem ser modulados pelos hormônios gonadais administrados em fêmeas OVX, onde a P gerou maiores resultados. Esses achados revelam ações adicionais dos esteróides sexuais na AMePD e novas características na morfologia neuronal em ratos machos e fêmeas adultos, complementando e avançando a literatura atual com resultados que ampliam o número de ações dos hormônios gonadais nesta área telencefálica de ratos.pt_BR
dc.description.abstractIn rats, the posterodorsal medial amygdala (MePD) is a sexually dimorphic area and it is implicated in the control of reproduction. The male and female gonadal hormones have an important epigenetic role upon neuro-glial morphology and function in the rat MePD. In this work two great unpublished and relevant topics in the literature for the understanding of the cellular bases of functioning of the MePD were discovered: (1) the NADPH-diaphorase (NADPH-d) activity and the number of NADPH-d-positive neurons of adult males, females across the estrous cycle (diestrus, proestrus, estrus, and metaestrus) or following ovariectomy (OVX) and substitutive therapy consisting of vehicle/oil (control), estradiol benzoate alone (EB), estradiol benzoate and progesterone (EB+P), or progesterone alone (P), and, (2) the modification of the somatic and nuclear from MePD neurons of OVXed female rats submitted to the same hormonal therapy, aiming to reveal possible influence of sex steroids in these morphological parameters. The NADPH-d histochemical technique was used to study the regional and neuronal NADPH-d activity, followed by a semi-quantitative analysis using optical densitometry. All animals were submitted to the same experimental protocol. Males showed a higher regional and neuronal optical density when compared to females across the estrous cycle, with the exception of the diestrus phase (P < 0.01). No differences were found in these two parameters during the ovarian cycle (P > 0.05). As for the number of NADPH-d-positive neurons, there were no statistically significant differences found among males and cycling females (P > 0.05). Since the gonadal hormones are able to provide supra-physiologic answers different from those observed during the normal ovarian cycle in the MePD, the effects of the OVX were tested also in these parameters. No statistically significant difference was found in the regional optical density, in the neuronal optical density, or in the number of NADPH-d positive neurons when comparing the data from OVXed females treated with vehicle or the three different hormonal therapies (P = 0.07, P = 0.18, and P = 0.95, respectively). To measure the somatic and nuclear volumes from MePD neurons, one-week following ovariectomy and at the end of hormonal treatments, brains were cut to semithin sections (1 mm) and stained with 1% toluidine blue for stereological estimations, carried out using the Cavalieri method and the technique of point counting. Both the somatic and neuronal volume showed a statistically significant difference when comparing the data among OVXed females treated with vehicle (V), EB, EB+P or P. This is, OVX with EB+P treated females had higher mean somatic and nuclear volume when compared to V (P < 0.01) or with EB (P < 0.01). Also, OVXed females treated with P showed larger mean somatic and nuclear volume when compared to V (P < 0.05). Results suggest that NADPH-d activity in the rat MePD is different between sexes but in females is not affected by changing levels of circulating gonadal hormones in physiological or supraphysiological conditions. Moreover, somatic and nuclear volume can be modulated by substitutive ovarian hormones administered to OVXed females, for which P can lead to higher results. These findings reveal additional actions of the sexual steroids in the MePD and new features for the neuronal morphology in adult male and female rats, complementing and advancing the current literature with results that enlarge the number of actions of gonadal hormones in this telencephalic area of rats.en
