dc.description.abstractBacteriocinas são peptídeos com atividade bactericida ou bacteriostática para espécies sensíveis ao produto bacteriano. Estas substâncias são compostos heterogêneos em relação à massa molecular, propriedades bioquímicas, espectro inibitório e mecanismos de ação. Apesar dos trabalhos realizados quanto à biodiversidade da Amazônia, poucos estudos são desenvolvidos com o objetivo de investigar a complexidade genética e o potencial biotecnológico desta região, principalmente quando se trata de micro-organismos. Neste âmbito, o presente trabalho teve como objetivo verificar a presença de genes de bacteriocinas produzidas por diferentes linhagens de Bacillus isoladas de intestino de peixes da Região Amazônica, Brasil. Os PCRs mostraram que as cinco linhagens testadas apresentam o gene funcional codificante de malonil Coa transacilase (ituD), para o gene codificador da pré-subtilosina A (sboA) e para o gene codificador da pré-subtilina (spaS). Quatro isolados possuem também o suposto gene terminador transcricional (sfp). A inibição do crescimento de fungos fitopatogênicos caracterizou a atividade antimicótica dos isolados. A caracterização da atividade biossurfactante foi realizada pela presença de zonas de hemólise ao redor de cada colônia e atividade emulsificante. Análises realizadas por MALDI-TOF confirmam a produção de surfactina, iturina A, subtilina, subtilosina A e isoformas. Ao comparar as linhagens de Bacillus Amazônicos com Bacillus subtilis ATCC 19659 em relação ao nível de expressão de sboA e ituD, observou-se, de forma geral, um nível de expressão de sboA pelos isolados Amazônicos inferior a B. subtillis ATCC 19659. Em contraste, quando os níveis de ituD foram avaliados, percebeu-se que as linhagens Amazônicas exibiram altos níveis de expressão deste gene. Acredita-se que independente do controle regulatório a que estão subordinados os operons de iturina A e subtilosina A nas condições de estresse utilizadas por este estudo, os isolados de Bacillus possuem um nível basal de expressão diferenciado entre si e em relação à B. subtilis ATCC 19659.pt_BR
dc.description.abstractBacteriocins are peptides with bactericidal or bacteriostatic activity for species closely related to bacterial product. These substances are heterogeneous compounds in relation to molecular weight, biochemical properties, inhibitory spectrum and mechanisms of action. Despite the work done on the biodiversity of the Amazon, few studies were developed with the objective of investigating the genetic complexity and biotechnology potential in this region, especially when it comes to microorganisms. In this context, this study aimed to verify the presence of genes of bacteriocins produced by five strains of Bacillus isolated from aquatic environment of the Amazon region, Brazil. The PCRs showed that strains tested exhibit potential for the functional gene encoding for malonyl CoA transacilase (ituD), for the gene encoding prosubtilosin A (sboA), and the gene encoding prosubtilin (spaS). Four isolates also have potential for the putative transcriptional terminator gene (sfp). Inhibiting the growth of pathogenic fungi characterizes the antimycotic activity of the isolates. The characterization of biosurfactant activity is undertaken by the presence of zones of hemolysis around each colony, as well the emulsifying activity. Analyses carried out by MALDI-TOF mass spectrometry confirmed the production of surfactin, iturin A subtilin, subtilosin A and isoforms. By comparing Bacillus strains isolated from the Amazon region with Bacillus subtilis ATCC 19659 on the level of sboA and ituD expression, there was, overall, a level of sboA expression of the isolated Amazonian less than B. subtilis ATCC 19659. In contrast, when levels of ituD were evaluated, it was noticed that the Amazonian strains exhibited high levels of expression of this gene. It is believed that regardless of regulatory control that are the subject of iturin A and subtilosin A operons in stress conditions used in this study, isolates of Bacillus have a baseline level of expression is different among themselves and in relation to B. subtilis ATCC 19659.en
