dc.description.abstractNeste trabalho foram desenvolvidas cinco simulações: Moricina no vácuo; moricina em meio fisiológico (água e íons); membrana em meio fisiológico; 1 moricina e 6 moricinas no interior da membrana. O objetivo é avaliar as interações antibiótico – membrana. Para esse estudo foi escolhido um peptídeo catiônico com atividade bactericida já constatada. A membrana adotada é baseada na composição da membrana celular da bactéria E. Coli. A metodologia utilizada foi a Dinâmica Molecular. Os sistemas foram construídos com o software VMD e os cálculos foram desenvolvidos com o software NAMD2. O Campo de Força para os lipídios, os íons e a moricina foi o CHARMM27 e o modelo para água foi TIP3P. O ensemble adotado foi o NpT, com uma pressão de 1 atm e temperatura de 310 K, por ser o mais coerente com o sistema fisiológico. A moricina é um peptídeo α-hélice e por isso o modelo de atuação aplicado foi barrel stave. Em presença do peptídeo se observou a perturbação da estrutura da membrana, o que valida a metodologia. No sistema contendo 6 moricinas o poro formado possibilitou o escoamento de água e íons através da bicamada lipídica, desfazendo o gradiente de concentrações essencial à manutenção da célula bacteriana.pt_BR
dc.description.abstractIn the present dissertation, five systems have been treated by molecular dynamics computer simulations: isolated moricine, moricine in physiological solution, membrane in physiological solution, a single moricine molecule inserted into the membrane, and an ion channel consisted of six moricine molecules inserted into the membrane. The moricine molecule represents a cationic peptide with known antibiotic activity. The membrane model was chosen to mimic the composition of the celular membrane of E.Coli. Our studies were directed towards the interaction between the antibiotic moricine and the membrane. All the systems were constructed using the VMD software. The simulations have been performed with the NAMD2 package adopting the CHARMM27 force field to the lipides, ions, and the moricine molecule. Water was described by the TIP3P model. The simulations were carried out within the NpT ensemble corresponding to physiological conditions (pressure of 1 atm and a temperature of 310 K). Moricine is a peptide with alpha-helix treated as a barrel stave. The adopted methodology demonstrated a perturbation of the membrane's structure in the presence of the peptide. The system containing six moricine molecules forming a pore within the membrane exhibited a flux of water molecules and ions passing the membrane indicating the antibiotic activity of moricine.en
