dc.description.abstractA identificação de alelos e genótipos associados a características reprodutivas, somente foi possível a partir do desenvolvimento e da localização de marcadores moleculares polimórficos no genoma, especialmente os short tandem repeated (STRs) e os single nucleotide polymorphysm (SNPs). A termografia infravermelho tem sido amplamente empregada como um método não invasivo na determinação da temperatura da superfície corporal, mostrando-se uma técnica promissora na avaliação da adaptação a efeitos ambientais. Portanto, O objetivo geral deste estudo foi identificar marcadores moleculares relacionados à fertilidade de touros e utilizar a termografia infravermelho na avaliação adicional ao exame da aptidão reprodutiva desses animais, medindo-se a temperatura da superfície escrotal. A amostra constou de 87 touros submetidos à analise e acompanhamento da mensuração do perímetro escrotal (PE), dos 7 aos 24 meses de idade. Esses touros foram analisados andrologicamente aos 24 meses pela Classificação Andrológica por Pontos (CAP). Subsequentemente, 17 reprodutores da amostra tiveram seu PE medido e foram examinados e pontuados pelo CAP aos 28, 32 e 36 meses de idade. Não foi verificada associação entre os marcadores STRs BMS3004, HEL5, AFZ1, IDVGA51 e ILSTS002 e a desempenho reprodutivo em ambos os experimentos. O SNP FSHR mostrou significativa relação com o genótipo GG para menor PE aos 24 meses. Os genótipos CG e GG foram significativos para identificação do PE inferior a 29 cm e o genótipo CC para maior pontuação no CAP. Durante o experimento II no SNP FSHR foi encontrada relação ao maior PE aos 7meses, 24meses, 28 meses, 32 meses e 36 meses de idade com o genótipo CC, mas nada foi encontrado de relação com o CAP em ambos os marcadores. Em conclusão, observamos uma forte associação do SNP FSHR com o genótipo GG para PE reduzido e CAP superior no genótipo CC, e a possibilidade de quando encontrado os genótipos CG e GG, encontrar um PE inferior a dois desvios a menos da média da raça. Durante o experimento II também foram avaliados os efeitos sazonais do ambiente sobre a qualidade do sêmen em reprodutores, utilizando-se termografia infravermelho. Variações de gradiente de temperatura escrotal foram significativamente maiores no outono (4.5°C) e inverno (4.0°C) do que na primavera (2.9°C) e verão (0.9°C) (P<0,05). Temperaturas do globo ocular foram mais baixas no inverno (27.6°C) e outono (26.8°C) em relação ao verão (33.9°C) e primavera (31.1°C) (P<0,05). Motilidade do sêmen,movimento de massa e vigor diminuíram no verão em comparação com as outras estações. Concluiu-se que a termografia infravermelho pode ser adotada como um método indireto para avaliar gradiente testicular e suas consequências aos aspectos físicos seminais.pt_BR
dc.description.abstractThe identification of alleles and genotype associated with these features was only possible through the development and localization of polymorphic molecular markers in the bovine genome, especially the short tandem repeated (STR) and single nucleotide polymorphysm (SNP). Infrared thermography has been widely used as a noninvasive method for determining the temperature of the body surface. This tecnhique shows a promising method to assess the adaptation of environmental effects. Therefore, the aim of this study was to identify molecular markers that are linked to bulls fertility. We also evaluted the uses of infrared thermography to evaluate the reproductive performance of bulls, measuring the surface temperature of the scrotum. Samples were collected from of 87 bulls submitted to analysis from 7 to 36 mo of age measurement of scrotal circumference and analyzed by BBSE. Subsequently, 17 bulls the previous sample measured and had their PE and was examined by BBSE at 28, 32 and 36 mo old. There was no significant association between markers BMS3004 STRs, HEL5, AFZ1, IDVGA51 ILSTS002 and reproductive performance in both experiments. The FSHR SNP showed a significant association with the GG genotype for low SC at 24 mo. The CG and GG genotypes were different for identification of SC less than 29 cm and CC genotype for highest score the BBSE. During the second experiment, in relation to the SNP FSHR at 7, 24, 28, 32 and 36 mo of age were found different with the CC genotype, but no correlation was found in relation to the BBSE in both markers. In conclusion, we observed a strong association of FSHR SNP genotype GG higher for SC and BBSE reduced the possibility of CC genotype and when found the CG and GG genotypes. During the second experiment also, we evaluated the seasonal effects of the environment on semen quality in breeding using infrared thermography images data that were collected using an infrared camera. Changes in temperature scrotal gradient were significantly higher in autumn (4.5° C) , winter (4.0°C) and spring (2.9°C) compared to the summer (0.9°C) (P < 0.05). Ocular temperatures were lower in winter (27.6°C) and autumn (26.8°C) compared to summer (33.9°C) and spring (31.1°C) (P < 0.05). Sperm motility, mass motion and vigor decreased in the summer compared with other seasons. It was concluded that infrared thermography can be used as an indirect method to assess testicular gradient and its consequences to the seminal physical aspects.en
