dc.description.abstractO degrane é uma das principais características que tornam o arroz vermelho daninho. A compreensão da regulação do degrane em arroz vermelho permitirá o desenvolvimento de procedimentos de biotecnologia que reduzam os problemas causados por esta planta daninha. O objetivo geral deste trabalho foi determinar a variabilidade e a regulação gênica do degrane de sementes em arroz vermelho. Os objetivos específicos foram i) caracterizar fenotipicamente o degrane das sementes em cultivares de arroz, genótipos de arroz silvestre, e ecótipos de arroz vermelho; ii) caracterizar a expressão dos genes conhecidos relacionados ao degrane em populações contrastantes; iii) analisar a variabilidade nucleotídica de genes relacionados direta e indiretamente ao degrane nestas populações; iv) identificar novas sequências gênicas expressas diferencialmente em ecótipos com diferentes níveis de degrane. Os genótipos avaliados apresentaram elevada variabilidade do degrane que variou de 20 a 270 gf. O gene qSH1 comumente relacionado com o degrane não possui efeito nos genótipos avaliados. A expressão relativa dos genes OsCPL1 e OsXTH8 apresentou uma relação direta com o nível de degrane. Já a expressão relativa do gene OsCel9D apresentou uma relação inversa aos níveis de degrane. A variabilidade nucleotídica do gene Os08g0512400 a 1271 bases upstream mostrou que os genótipos com o nucleotídeo T possuem em geral elevado degrane, enquanto que os genótipos com o nucleotídeo A apresentam baixo degrane. Além disso, a variação nucleotídica do exon 5 do gene Os01g0849100 apresentou dois SNPs, nas posições 2981 e 3057, que estão relacionados ao degrane. A metodologia SSH identificou 154 clones diferencialmente expressos que podem estar relacionados ao degrane em arroz. Destes clones, 61% apresentam funções conhecidas relacionadas ao processamento e armazenamento da informação, sinalização, processos celulares e metabolismo. Além de genes conhecidos como OsCPL1, os genes OsXTH8, OsCel9D, Os08g0512400 e Os01g0849100 também estão relacionados ao degrane de sementes em arroz.pt_BR
dc.description.abstractSeed shattering is one of the main traits that make the red rice a weed. Better understanding of seed shattering regulation in red rice can be used to develop biotechnological procedures to reduce the problems of this weed. The main objective of this study was to determine the variability and genetic regulation of seed shattering in red rice. The specific objectives were i) to characterize the seed shattering in rice cultivars, wild rice and red rice ecotypes; ii) to characterize the expression of known genes related to seed shattering in contrasting rice genotypes; iii) to analyze the nucleotide variability of genes directly and indirectly related to seed shattering in these genotypes, and iv) identify new gene sequences differentially expressed in ecotypes with different levels of seed shattering. The evaluated genotypes presented large variability of seed shattering, which ranged from 20 to 270 gf. The gene qSH1, commonly related to seed shattering had no effect on the evaluated genotypes. The expression of the genes OsCPL1 and OsXTH8 was directly related with seed shattering. Otherwise, the expression of the gene OsCel9D was inversely related with seed shattering. The gene Os08g0512400 was polymorphic at 1271 bases upstream, where, in general, the genotypes with the T nucleotide had high seed shattering, and with the A nucleotide had low seed shattering. In addition, the exon 5 of the gene Os01g0849100 presented two SNPs at positions 2981 and 3057, which may be related to seed shattering. The SSH study identified 154 clones genes differentially expressed that may be related to seed shattering. The sequencing and analysis of these clones indicated that 61% of then have known functions related to processing and storage of information, signaling, cellular processes and metabolism. The variability of seed shattering in red rice is related with several genes. In addition to the know gene OsCPL1, the genes OsXTH8, OsCel9D, Os08g0512400 and Os01g0849100 are also related to seed shattering in rice.en
